Animals actively use at least half of the genome

چکیده
10 One central goal of genome biology is to understand how the usage of the genome differs between 11 organisms. Our knowledge of genome composition, needed for downstream inferences, is critically 12 dependent on gene annotations, yet problems associated with gene annotation and assembly errors are 13 usually ignored in comparative genomics. Here we analyze the genomes of 68 species across all animal 14 groups and some single-cell eukaryotes for general trends in genome usage and composition, taking into 15 account problems of gene annotation. We show that, regardless of genome size, the ratio of introns to 16 intergenic sequence is comparable across essentially all animals, with nearly all deviations dominated by 17 increased intergenic sequence. Genomes of model organisms have ratios much closer to 1:1, suggesting that 18 the majority of published genomes of non-model organisms are underannotated and consequently omit 19 substantial numbers of genes, with likely negative impact on evolutionary interpretations. Finally, our 20 results also indicate that most animals transcribe half or more of their genomes arguing against differences 21 in genome usage between animal groups, and also suggesting that the transcribed portion is more 22 dependent on genome size than previously thought. 23

طرح تحلیل تعدادی از mRNA های مادری مخصوص اووسیت در جنین تک سلولی موش
Introduction & Objective: During oogenesis, mRNA is actively transcribed and accumulated in the growing oocytes, and then the transcription stops. Transcription silencing will continue during early embryonic stages at least up to the time when the embryonic genome is activated. Thus the earliest stages of embryogenesis in mammals and other animal species are depending on stored maternal RNAs an...
متن کاملInvestigation of DNA Integration into Reproductive Organs Following Intramuscular Injection of DNA in Mice
Background: DNA immunization with plasmid DNA encoding bacterial, viral, parasitic, and tumor antigens has been reported to trigger protective immunity. The use of plasmid DNA vaccinations against many diseases has produced promising results in animal and human clinical trials; however, safety concerns about the use of DNA vaccines exist, such as the possibility of integration into the host gen...

متن کاملI-49: Human Y Chromosome ProteomeProject
The success of the Human Genome Project (HGP) has provided a blueprint for the approximately 20,000 gene-encoded proteins potentially active in all of the hundreds of cell types that make up the human body. Yet we still have limited knowledge about a majority of the gene-encoded proteins which are the “building blocks of life” and “cellular machinery”. It is estimated that for nearly half of th...

Background: PhiC31 integrase system provides a new platform in various felid of research, mainly in gene therapy and creation of transgenic animals. This system enables integration of exogenous DNA into preferred locations in mammalian genomes, which results in robust, long-term expression of the integrated transgene. Objectives: Identification of a novel pseudo attP site. Materials and Methods...
